| Research Abstract |
ras関連GTP結合タンパク質は細胞内の色々な調節中心的な働きをしている。例えば.成長,分化,細胞骨格の構成,細胞輸送など。こうした働きは、GTP結合タンパク質のC末端側が.大切であることが知られている。こうした重要性にも関わらず、植物における.このタンパク質の研究は進んでいない。本実験の目的は、GTP結合タンパク質遺伝子の発現と、そのタンパク質の細胞内の局在を調べることである。
1.コムギジェノミックライブラリーを作った。wgp7をコードするクローンをいくつか単離した。その塩基配列を決定中である。
2.wgp7をセンス及びアンチセンス方向にタバコに組みこんだ。幼植物体コントロールに比べて、形態状の変化が見られなかった。
3.wgp7のC末端側45アミノ酸をGUSに結合した、ベクターを作った。このベクターをタバコに導入し、wgp7タンパク質の細胞内局在を調べる予定である。
