| Research Abstract |
悪性黒色腫に高い特異性を有するとされるモノクローナル抗体・HMB-45を用い、我々は日本人皮膚腫瘍(特に色素細胞系腫瘍)においてその特異性および臨床応用につきすでに成果を発表した(菊地 新他、日皮会誌 102:577-581,1992)。HMB-45にて認識されるHMB-45抗原は、最近になり悪性黒色腫細胞のみならず発達中の色素細胞にも発現されていることが明らかになったが、その局在および本体を知ることは悪性黒色腫細胞の性質を研究する上で必須である。今回我々は、まずチロジナーゼ陰性眼皮膚白皮症および正常胎児におけるHMB-45抗原の発現とその微細構造における局在につき検討した結果、これらは胎生初期より存在しかつ量的相違こそあれ全身のあらゆる皮膚に分布し、未熟なメラノゾーム上に局在していることが金コロイドを用いたpost-enbedding法免疫電子顕微鏡にて確認された(Kikuchi A et al.Immunohistochemical and ultra-structural studies of the development of melanocytes in the fetal skin in the second trimester,The European Society for Dermatological Research 23rd Meeting.April 3-6 1993.)。またこれら胎児のみならず、さまざまな種類の色素細胞[メラノーマ細胞樹立株(メラニン産生有り・無し)、悪性黒色腫患者生標本(melanotic,amelanotic)、チロジナーゼ陰性眼皮膚白皮症および正常小児]においてその局在を検討したところ、HMB-45は主として未熟なメラノゾーム上に局在しするものの、成熟したIII型およびIV型メラノゾーム上にも発現されており、これらはそれぞれの条件で量的にかなり差異があることが判明した(Kikuchi A et al.Ultrastructural localization of HMB-45 antigen in the fetal melanocytes and melanoma cells,The Second Tricontinental Meeting of JSID,SID and ESDR.October 28-31,1993.)。
