Research Abstract

Since this program for functional analysis of tumor suppressor genes has started in the US several years ago, we plan to continue and explore the scientific communication as a US-Japan collaborative research. We have been establishing the system that allows to analyze functional aspects of the tumor suppressor genes, such as the RB,p53 and NF1 genes, by stably transfecting the wild-type gene into cancer cells whose genes are mutated. Comparing the phenotype of the parental and transfected-cells, we elucidated the functional role of each gene, especially at the cellular level. We also performed experiments controling the expression levels of transfected genes that causes drastic phenotypic changes, such as apoptosis and differentiation. Our experimental model should provide a clue to the possible development of a new cancer therapy.
