Research Abstract	心臓の肥大・拡張モデルの確立と心機能評価 (1)家兎の頚部動脈と静脈間に側々吻合路を作製し、容量負荷心を作製した。短絡血流量を安静時頚動脈血流量の4倍に設定したところ、慢性期に著明な心臓の拡大と心筋の肥大を観察することができた。すなわち、動静脈短絡を作製して12週目に心臓の形態計測を行ったところ、Sham手術例に比較して右室は65%,左室は66%の重量増加と心内容積の増大を認めた。心機能の変化については、カテ先血圧形を用いて左室の内圧を測定し、dp/dtを心収縮能の指標とした。短絡路形成による容量負荷12週において,Ca^<2+>やβ受容体刺激に対する反応性の低下を認めた。すなわち、心肥大の過程においてβ受容体のdown regulationが示唆された。Ca^<2+>感受性の低下が何時生じたかを明らかにする為経時的検討を行った。 (2)右室の圧負荷モデルについてはモノクロタリンの腹腔内注射によって作製することができた。TxA_2受容体拮抗剤に対する反応からモノクロタリン注入初期における血小板凝集亢進が病態の形成に関与していたものと思われた。 (3)右室と左室の相互干渉に関する検討 β受容体刺激とCa^<2+>拮抗剤の右心室機能と左心室機能に及ぼす効果を検討した。
