| Research Abstract |
D-myb遺伝子はX染色体13E-Fに位置する。EMS処理によりX染色体劣性致死変異体を約1,000系統分離した。一方P因子によりD-myb遺伝子を第2あるいは第3染色体に組み込んだautosome transformantを作製し、上記のX染色体劣勢致死変異体と掛け合わせ、致死性が相補されるものをスクリーニングした。その結果1系統(el2507)が分離され、遺伝的マッピングより変異はX染色体上10Eと15Fの間に位置することが示された。現在、この系統のD-myb遺伝子のDNA配列を決定しつつある。cAMP経路に関与する転写因子の変異体を分離するため、P因子を用いて得られたエンハンサートラップ系統を作製し、β-ガラクトシダーゼの発現が特に脳で高い系統を得た。P因子が挿入した遺伝子のcDNAクローニングを行ない、塩基配列を決定した結果、この遺伝子にコードされる蛋白質はマウスCBPのCREB結合ドメインと高いホモロジーを有していた。そして実際にin vitro翻訳系で産生したD-CREB-BはPKAでリン酸化した時にこの遺伝子産物に結合することから、この遺伝子がDrosophila CBPをコードすることが明かとなった。D-CBP遺伝子にP因子が挿入した系統は劣性致死であり、germ lineモザイク解析の結果D-CBPは頭部の形態形成や中枢神経系のlongitudinal connectivesとcommisuresの形成に必要であることが示された。
