| Research Abstract |
我々は早期活性化マーカー分子CD69を用いてポジティブセレクションの起こりつつある細胞群を同定した。ポジティブセレクションが起こる際とネガティブセレクションが起こる際に誘起されるシグナル伝達を次のような方法で解析しポジティブセレクションに特有のシグナル伝達機構の解明を目的とする。(1)細胞表面のTCRやCD69の量をフローサイトメトリーで、(2)チロシンキナーゼの活性化を抗チロシンリン酸抗体を使ったイムノブロッィング法で、(3)〔Ca^<2+>〕iの変化をコンピュータ画像解析装置(国立予防衛生研究所の鈴木和男博士の装置を利用)を用いて、(4)RAG-1、RAG-2といったT細胞のセレクションとともに発現の変化する分子のmRNAをノザンブロッティング法で解析した。免疫抑制剤であるFK506やサイクロスポリンAを用いてポジティブセレクションやネガティブセレクションのシグナル伝達系を解析したところ、胸腺内でのアポトーシスを伴うネガティブセレクションにはカルシニューリンの活性化は必要ないことがわかった。さらに、ポジティブセレクションついては、そのステップの最初の段階でカルシニューリンの活性化が必須であることがわかった。
さらには、末梢の成熟T細胞に誘導されるアポトーシスはFK506やサイクロスポリンAで完全に抑制されるため胸腺細胞でのアポトーシスと成熟T細胞でのアポトーシスは違ったメカニズムで起こっていることがわかった。
