Research Abstract	1)大腸がんのがん抑制遺伝子APCの産物がDrosophilaのがん抑制遺伝子DLGのヒトホモログの産物と複合体を形成することを見出し、この複合体形成がAPCによる細胞周期制御に重要であることを示唆する実験結果を得た。2)DLGの機能をさらに明らかにするためにDLG結合蛋白質を検索し、guanylate kinaseドメインに結合する蛋白質をコードする新規遺伝子DAP-1、DAP-2、DAP-3を単離し機能を解析した。この結果からguanylate kinaseとアミノ酸配列のホモロジーがありながら酵素活性がないことから機能の不明であったこのドメインが蛋白質-蛋白質相互作用に関与するドメインであることが明らかになった。3)SH3ドメインについては分子内でguanylate kinaseドメインを含む領域と結合している可能性があることを見出した。4)DLGのリン酸化にCDC2とEGF受容体下流のキナーゼが関与している可能性が示唆された。5)APC蛋白質のarmadilloモチーフに結合する蛋白質を検索しSam68を含め三種類の蛋白質が結合することを見出した。Sam68は、Srcと結合しチロシンリン酸化をうけること、C.elegansのがん抑制遺伝子産物GLD-1のヒトホモログであることが知られている。6)armadilloに結合する蛋白質をコードする2種類の新規遺伝子を見出し、これらがarmadilloのシグナル伝達経路上で機能していることを見出した。
