DYNAMIC ASPECTS CONCERNING SPECIFIC MOLECULAR RECOGNITION PROCESSES AT LIPID MEMBRANE INTERFACES WERE INVESTIGATED BY MAKING USE OF VIBRATION SPECTROSCOPY (SURFACE-ENHANCED RAMAN SPECTROSCOPY (SERS) AND FOURIER-TRANSFORM INFRA RED (FTIR) SPECTROSCOPY,FOR EXAMPLE), MULTIPLE INTERNAL REFLECTION FLUORESCENCE METHOD (MIRF), AND MICROSCOPIC METHOD. (1) CYTOCHROME C BOUND TO SELF-ASSEMBLED MONOLAYERS (SAM) ON SILVER WAS STUDIED BY SERS,AND IT WAS CONCLUDED THAT SILVER MODIFIED WITH A SAM IS A SUITABLE SERS-ACTIVE SUBSTRATE TO ANALYZE THE STRUCTURE OF PROTEINS ON BIOMIMETIC MEMBRANE SURFACES WITHOUT DENATURATION. (2) NOVEL AMPHIPHILES WHICH CONTAIN GALACTOSE RESIDUES WERE PREPARED BY TELOMERIZATION OF GALACTOSE-CARRYING VINYL MONOMER USING A LIPOPHILIC RADICAL INITIATOR.THE GALACTOSE-CARRYING AMPHIPHILES INCORPORATED IN A LIPOSOME WAS EASILY RECOGNIZED BY A LECTIN FROM Ricinus communis AND GALACTOSE OXIDASE.THE RECOGNITION WAS LARGELY AFFECTED BY THE DEGREE OF POLYMERIZATION AND THE SURFACE DENSITY OF THE AMPHIPHILE. (3) BINDING PROCESSES OF ANTIBODY TO A HAPTEN-DOPED LIPID MONOLAYERS WERE FOLLOWED BY THE MULTIPLE INTERNAL REFLECTION FLUORESCENCE METHOD.THE INITIAL RATE OF BINDING OF THE ANTIBODY INCREASED AT A LOW SURFACE DENSITY OF HAPTENS IN THE LIPID MONOLAYER,AND LEVELED OFF AT A MODERATE-TO-HIGH HAPTEN DENSITY.THE LONG SPACER BETWEEN THE HAPTEN GROUP AND THE LIPID LAYER PROMOTED THE ACCESS OF ANTIBODIES TO THE HAPTENS.
