Research Abstract

For understanding the evolutionary ralationships between gonadotropins (LH and FSH) and their receptors, we attempted to charatcterized the extracellulara domain of the receptors, which is thought to be a key region of hormone binding, in nonmammalian species, and to compare the information to that of the known mammalian data. By using polymerase chain reaction (PCR) and reverse transcription (RT)-PCR,parts of the quail and newt FSH receptor cDNAs were amplified. The homology of predicted peptide of the quail and newt FSH receptors was about 70% and 67% of those of mammalian FSH receptors, resepectively. We also charactreized cDNA of the quail LH receptor. The homology of both cDNA and predicted peptide was about 70% of those of mammalian LH receptors. These data suggetsts that the ligand binding domain of receptro is more conserved than the ligand molecules during their evolution. To reinforce this hypothesis, molecular characterization of FSH and LH receptors in a wider range of vertebrate classes is needed.
