| Research Abstract |
レクチン-プロテアーゼ(MBP-MASP)複合体による補体の活性化は、認識機構を持たない非特異的な活性化の第二経路や、抗原抗体を認識してC1に始まる古典的経路とは異なる新たな補体活性化経路(レクチン経路)の存在すを意味する。そこで、本研究はレクチン経路の解析と生体での役割を明らかにする事を目的とし、本年度は次のような結果が得られた。
1、MBP-MASPによる活性化経路の解析、特にC3分解の検討:MASPは、血清中では前駆体として存在し、MBPがそのリガンドに結合すると活性化されることと、それ自身でautoactivation を起こすことが判明した。また、MASPは直接、C3を活性化し、古典的経路より、古くから存在していたことが推察される(Immunobiol.,1995)。
2、当初計画していたリコンビナントMASPを得ることはできなかったが、ハーバード大学のEzekowitz博士との共同研究の結果、リコンビナントMBPもMASPを介して補体系を活性化することが明らかにされ、また、変異MBPはMASPを結合できず、補体の活性化も起こらないことが判明した(Biochem.J.,1995)。
3、ヒトMASP遺伝子解析の結果、セリンプロテアーゼドメインは、C1sと異なり、エクソン、イントロン構造をとることが判明した(論文投稿中)。
