Research Abstract	本年度の研究により正常肝の部分切除後には第3,7,14日目にIII,IV型コラーゲンのmRNAの発現が増強することが判明した。すなわち,これら3つのピーク時にはその他の時期に比べて2-3倍のmRMAの発現が見られた。また,Brdu染色によりDNAの合成を経済的に調べると,門脈域より中心静脈にむかって染色されることが判明した。このことは肝部分切除後の肝再生が一度に起こるのではなく波の如く,門脈域より派生するものと考えられる。この実験系にコラーゲンの合成酵素(プロリルハイドロキシラーゼ)阻害剤を加えると部分肝切除後14日目には通常,もとの大きさの肝臓になるところが抑制され,Brdu染色も有意に抑制されDNA合成が抑制されていることがわかった。これらのことから,コラーゲンを初めとする細胞外マトリックスは肝細胞の再生に必要なものであることがわかった。さらに,本年度の研究より肝部分切除後には肝障害で重要な役割をになっているTumor necrosis factor(TNF-α)のmRNA発現が早期(24-48時間後)に増加することがわかり,TNF-αの抑制剤(2E-3-5-2,3-dimethoxy-6-methyl-1,4-benzoquinoyl-2-nonyl-2-propenoic acid,E3330)を加えることにより肝再生が抑制される,予備実験結果をえており,現在本試験ならびに細胞外マトリックスとの相互関連を検討中である。
