Research Abstract	アルギオテンシンII(Ang II)は特異的受容体を介して強力な昇圧作用や細胞増殖作用を発揮する。AngII type1a受容体(AT1a-R)は心血管系では豊富に発現し神経細胞ではその発現は低く抑制されている。私達はAT1-R遺伝子の上流域を解析しこの細胞特異的発現に関与するcis調節領域の同定および病態心での発現調節を検討した。陰性cis調節配列のマッピングゲル移動度シフト法及びフットプリント法にて陰性cis配列は上流-456から-442に存在するA+T rich配列である事を明らかにした。同様のA+T rich配列は他の遺伝子においてsilencerとして働く事が報告されている。サウスウエスタン法においてこの配列に結合する核蛋白は約53KDaの分子量を持ち神経細胞に特異的に発現する事が明らかになった。糖質コルチコイド応答領域 (GRE) の同定 CAT reporter 遺伝子及びゲル移動度シフト法によりGREは上流-771から-756に存在する事が明らかになった。この配列は典型的GREと3bp mismatchであったが糖質コルチコイドにより転写を約3倍誘導し、特異的核蛋白の結合も確認された。
