Research Abstract

In psoriasis and sterile pustulosis, chemotactic peptides consisting of complement fragments and chemokines (IL-8 and Gro-alpha) are demonstrable in lesional skin. They are presumed to play an important role in the induction of leukocyte chemotaxis from the dermal papillae (squirting papillae). C3, the key factor in the complement system is produced by keratinocytes. We studied cytokine regulation of C3 production by cultured human keratinocytes with ELISA by adding IL-1 alpha, beta, -2,3,4,5,6,7,8,10, TNF-alpha INF-alpha and -gamma, and GMCSF to the cultures. Among them, INF-gamma and TNF-alpha induced the synthesis of C3 in a dose dependent manner. There was a remarkable synergistic effect between them. Further analysis of C3 production revealed that these cytokines augmented the C3 production by epidermal Keratinocytes via different signal transduction pathways.
