Research Abstract

In our previous study of glutaraldehyde stabilized human umbilical veins (GHUVs), we confirmed that the internal elastic lamina shows excellent biological compatibility in a longterm implantation study using dogs. In order to obtain asmall cariber arterial prosthesis with such property, we produced the elastin coated polyurethan graft (ECG), in which the flow surface was covered by a thin gelatin layr coated with heparin bounded alpha-elastin, and investigated the antithrombogenicity and the histocompatibility of the grafts by implantation study using dogs.
The ECGs (5 cm-long with an internal diameter of 6 mm) were implanted into the abdominal aorta of 10 mongrel dogs, and were retrieved at intervals of 24 hours to 6 months after grafting. Although heparin was administered at implantation and prior to sacrifice, neither antiplatelet agents nor anticoagulants were given during the observation period. All of the ECGs were patent at the time of removal. There were no thrombus formation on the flow surface of the graft obtained at 24 hours and one week, and neither immune reaction nor excessive foreingn body reaction was noted. Organization of the graft was almost complete at three months, and intimal hyperplasia at the anastomoses was not recognized in the grafts at 3 and 6 months.
These results indicated that the ECG shows a significant biological compatibility in the same as the GHUVs, and has a possibillity of application for small cariber arterial reconstraction.
