子宮体部の多段階発癌に関わる遺伝子群の解析とその遺伝子治療の検討

Research Abstract

子宮体部癌について、臨床検体63例、前癌病変である腺腫性増殖症8例、異型増殖症13例について、多段階発癌に関与する遺伝子群、特に癌原遺伝子(C-myc、C-erb-2、K-ras)、癌抑制遺伝子群(p53、Rb、DCC、APC、MTSなど)の変異を検索した。また一部の症例では未分化腺癌部や高分化腺癌部を個別に切り出し、それぞれのDNAを分析した。この結果は癌症例のうち80%に何らかの遺伝子変異が発見された。腺腫性病変では遺伝子変異は認めないが、異型増殖症では数%に遺伝子変異が検出できた。human papillomavirus ゲノムが約5%の子宮体癌症例の細胞DNAに組み込まれていることも示した。従って、平成6年度の結果により、子宮体癌の発症には、臨床病理学的多段階説と同様に遺伝子異常も多段階的に蓄積していることが臨床的にも示唆された。
