動脈硬化壁に蓄積する過酸化リポタンパク質認識モノクロナル抗体の開発

Research Abstract

動脈硬化壁内膜肥厚部に局在する泡沫細胞化形成に、過酸化リポタンパク質が重要な役割を果たしていると考えられている。我々はヒト動脈硬化巣のホモジェネートを抗原としてマウス(BALB/c)を感作し、過酸化リポタンパク質を認識するモノクロナル抗体産生株(FOHIa/DLH3株)を分離した。本モノクロナル抗体は、酸化LDLに対して強い,反応性を示す一方、未処理LDL、アセチルLDL、マロンジアルデヒドLDLとは反応しない。また、免疫染色によりヒト冠状動脈内膜肥厚部に局在する泡沫細胞を認識することが明らかになった。しかも、本抗体が認識する抗原決定部位は過酸化ホスファチジルコリンとペプチドの複合体の可能性が高い。これらの知見は、これまで動脈硬化巣に蓄積していることが示唆されてきた過酸化リポタンパク質の本体に一歩近づいたと考えている。
