Research Abstract

まず、本ラットの腎癌発生は放射線発癌および化学発癌に高感受性であることを明らかにした。この現象を利用して我々は、遺伝解析の為の,戻し交配ラットを短期間に多数,作成する方法を開発した。そして、原因遺伝子はラット染色体10番に局在することを明らかにした。また,ラット染色体10番上にあるDNAマーカーを用いることによって、腎癌細胞にはwild type alleleのloss(LOH)が高率に見られることがわかり,Knudsonの“two-hits"が適用され原因遺伝子は癌抑制遺伝子であることが考えられた。さらに、mappingを進め原因遺伝子は、ヒト結節性硬化症の原因遺伝子(TSC2;human chromosome 16p13.3)のrathomologueに強くリンクしていることを明らかにした。そしてついに、Tsc2遺伝子に本ラットに特異的なgerm line mutationを見いだした。興味あることに、本ラットには腎癌以外にも子宮腫瘍、脾臓腫瘍および下垂体腺腫が高率に発生することが明らかになった。
