Research Abstract	平成7年度は細胞間相互作用により誘起される細胞内カルシウム濃度の上昇を測定することを目的としていた。2種類以上の細胞を用い、より生理的反応に近い細胞間相互作用によって誘起される反応を測定することが目的であり、反応が散発的でゆっくりしているため、少なくとも1時間以上にわたって測定することが必要であった。そのため画像データ圧縮のプログラムの開発、簡略化(コンピュータープログラマ-に委託)、細胞の移動防止のための特殊な接着剤の使用と、移動した細胞を自動的に認識して画像処理するプログラムの開発(コンピュータープログラマ-に委託)、選別を行った。実際に抗原提示細胞とT細胞を用い、両細胞が充分細胞接着を起こした後、スーパー抗原を加えてT細胞を活性化させ、T細胞に誘起される細胞内カルシウム濃度の上昇を長時間モニターし、T細胞でオシレーションが起こっており、そのパターンが胸腺細胞と成熟T細胞で違うことがわかった。T細胞は、申請者たちの所有しているT細胞抗原レセプターのトランスジェニックマウスから調整し、T細胞の活性化がより効果的に起こるように工夫した。
