| Research Abstract |
DAF遺伝子導入トランスジェニックマウス及びHRF20遺伝子導入トランスジェニックマウスを樹立し、遺伝子発現をnortern hybridization及び免疫組織染色により確認した。これらマウスを交配し、DAF及びHRF20を同時に発現するトランスジェニックマウスを作成した。このマウスにヒト血清を静注し、ヒト血清による免疫反応がコントロールマウスに比べて著明に抑制されることを確認した。次にトランスジェニックマウス作成の技法及びベクターを用いて、DAF,HRF20及びMCP遺伝子導入トランスジェニックブタを作成するために、ブタ受精卵に対してこれら遺伝子を各々又は同時にマイクロインジェクションを行ない、現在トランスジェニックブタ作成を試行している。43頭のブタが出産され、そのうち2頭にヒト補体制御因子遺伝子導入を確認した。次に、異種抗原Galα1,3Gal epitopeを発現する酵素α1,3Galactosyl transferaseに対するマウス遺伝子をノックアウトするためにα1,3Galactosyl transferase遺伝子のsequenceを現在行なっている。
