Research Abstract	ラムダファージDNAの種々の部位にトランスポゾンを挿入し,PCRと磁気ビーズを用いる方法(Kasai et al.,1992)により,多量の塩基配列の生データを収集した。ラムダファーシDNAの全塩基配列はすでに既知なので,得られたデータについて各塩基の間の間隔(ピークの時間間隔)を測定し統計処理を行なうとともに,3′末端から10,20および30塩基の配列中のそれぞれの塩基の含量と電気泳動時のピーク間隔との関連を調べ,塩基配列と電気泳動の易動度の特色に関するデータベースを構築した。現在,このようにして作製したデータベースをもとにして,電気泳動データを塩基配列データに変換する,通常base-callerと呼ばれているコンピュータープログラムの作製・変更にとりかかり,従来のbase-callerに比較してどの程度の塩基配列決定の精度の向上が見られるかを解析すれための準備を進めている。さらに,これまでに得られている大腸菌の複製終結点付近の領域の多量のデータについて,この新しいbase-callerを用いての解析を進めるとともに,多数のデータの中からストップやコンプレッションのある塩基配列データを選び出し,確立した新しい方法で塩基配列決定を試み,その精度を検討する。それによって、現在進行中の実際のゲノム解析プロジェクトにこの方法を応用したときの評価を得られるようにしつつある。
