Research Abstract	マントル細胞リンパ腫を中心とする非ホジキンリンパ腫における cyclin D1の発現を分子生物学的、免疫組織科学的に評価し、RNAレベルでの過剰発現と抗 cyclinD1抗体による組織学的な核陽性所見が有為に相関することを明らかにし得た。さらに非ホジキンリンパ腫200例以上について抗cyclin D1抗体による染色所見を検討した結果、核陽性例がマントル細胞リンパ腫にほぼ限定され、細胞質における染色特性が節性リンパ腫と節外性リンパ腫では異なることを見出した。また、胃原発リンパ腫80例を粘膜関連リンパ組織型リンパ腫を中心として臨床病理学的に検討を加え、単に組織像だけではなく臨床病態においても明確な特徴を示す 4群に大別し得ることを明らかにした。研究の課程で、節外性リンパ腫に胚中心細胞由来のリンパ腫が極めて稀であり、胃リンパ腫の群別がp53,bcl-2の発現頻度と相関することを明らかにした。また、非ホジキンリンパ腫全体におけるCD56陽性リンパ腫の頻度を明らかにし、その多くがnatural killre細胞由来の可能性があり、一部の症例ではEpstein-Barr virusが関与することを示した。本研究により日常診断で分子病理学的腫瘍単位とも見做し得る"cyclin D1 over-expression lymphoma"の把握が可能となったが、その臨床病理学的意義は明らかではなく予後あるいは治療反応性を中心とした臨床病態の早急な解析の必要性が痛感された。一方、cyclin DI の発現の有無とその細胞内局在がリンパ腫組織型間で比較的多様であることが示されたが、その生物学的意義は今後検討されるべき問題と考えられた。また、胃原発リンパ腫群別の臨床病理学的妥当性を明らかにし得たことは、今後の分子病理学的、細菌学的研究における基盤となるものと考えられた。
