Research Abstract

Selenium is an essential trace element and the association with lipid metabolism has been suggested. Blood level of selenium has been used as an indicator of nutritional status of selenium. However, the level is dependent of chemical forms of selenium in the diet and plasma selenium can be divided into several groups including some selenoproteins and non-specific selenium-containing proteins.
In the present study, 1) we have estimated selenium intake by having foods classified into several food groups, 2) the distribution of selenium in human plasma has been investigated by an HPLC-ICP mass spectrometry method, and then 3) the effects of the forms of selenium in the diet and the distribution of selenium in plasma on the lipid concentrations in blood serum is now evaluated.
1) We have developed a new food intake frequency questionnaire and the results obtained from 50 women show that the questionnaire can detect 70.4% of daily selenium intake and among them 46.4% of selenium is from fishes, 45.5% is from animals (containing selenocysteine), and 8.0% is from cereals (mainly selenomethionine).
2) The chromatographic pattern of selenium in human plasma was investigated by an HPLC equipped with a multi-mode column (Asahipak GS-520 HQ) coupled directly to an ICP-MS.Four peaks of selenium were detected in the chromatogram. The first peak represents selenium contained in the high molecular weight fraction such as lipoproteins, and the second peak is thought to be selenoprotein-P.The retention time of the third peak was in accordance with that of albumin. The results show that the method can divide plasma selenium into three components.
