ウイルス性心筋炎における自己培養リンパ球注入による心筋障害機序の検討

Research Abstract

先に提出したプロトコールに従い研究を行った。すなわち、
実験I:Coxsackievirus B3とEncephalomyocarditis virusをBALB/Cマウスに接種し,ウイルス接種後約60日目にFicoll重層法にてspleenのリンパ球を得た。次に得られたTリンパ球を10%インターロイキン-2(IL-2)で培養した。
実験II:Coxsackievirus B3をBALB/Cマウスに接種し,ウイルス接種30日目に実験Iより得られたリンパ球(2×10^7個)を尾静脈より注入した。病理組織学的検索の結果,coxsackievirus B3感染マウスのspleen細胞を注入されたマウスでは,encephalomyocard:tis virus感染マウスのspleen細胞を注入されたマウスに比し,心筋炎の程度が増強していた。以上より,慢性心筋炎の病因に同一のウイルスで感作された細胞障害性T細胞が必要であることが判明した。現在は,リンパ球各サブセットの役割を検討中である。
