Research Abstract |
This study was designed to clarify whether basic fibroblast growth factor (bFGF) might increase regional myocardial blood flow (Qm) at the acutely infarcted myocardium in experimental animals. Methods : In 22 SD rats, the chest was opened and the left coronary artery was ligated. The chest was closed and bred for 4 weeks. Nonradioactive colored microsphere technique was used to measure Qm. In 11 rats, bFGF was not used while in the other 11 rats, bFGF 100mug was injected into the infarcted myocardium immediately after coronary occlusion. After 4 weeks, the heart was excised for the histological examinations. In 25 dogs, the left anterior descending coronary artery was ligated and bred for 4 weeks. Qm was measured using nonradioactive colored microsphere technique. In 11 dogs, bFGF 300mug was injected into the myocardium just before coronary occlusion, while in the other 14 dogs, bFGF was not given. Results : In rats, bFGF failed to change Qm of the infarcted myocardium as well as border zone and noninfarcted myocardium. Infarct size was not reduced in rats by bFGF.In dogs, Qm in control group and bFGF group were 24(]SY.+-.])4,63(]SY.+-.])10% respectively at the inner layr ; 23(]SY.+-.])5,100(]SY.+-.])20% at the middle layr and 74(]SY.+-.])7,130(]SY.+-.])16% at the outer layr. This study showed thatr bFGF increased Qm at the infarcted myocardium in dogs while bFGF had no effect in rats. Conclusion : Basic FGF had a significant effect to increase Qm at the infarcted myocardium in dogs. This may offer a new method for the treament of ischemic heart disease.
