| Research Abstract |
1.血友病A犬:血友病A保因犬の1匹が妊娠中で、現在出産準備中である。
2.イヌ第VIII因子・ヒト第VIII因子に対する抗第VIII因子同種抗体(インヒビター)およびモノクローナル抗体の交叉反応性:第VIII因子重鎖A2ドメイン認識インヒビターおよび数種の抗A2ドメインモノクローナル抗体はイヌ第VIII因子に対し反応性は見られなかった。同様に、第VIII因子軽鎖のN末端側に位置するA3ドメイン認識モノクローナル抗体もイヌ第VIII因子には反応しなかったが、軽鎖のC末端側のC2ドメインを認識するインヒビターやモノクローナル抗体は、ヒト第VIII因子と同程度にイヌ第VIII因子に反応した。従って、イヌ第VIII因子とヒト第VIII因子軽鎖C2ドメインの共通抗原性はきわめて高く、第VIII因子構造機能上本ドメインが重要であることが明らかになった。C2度目の重要性をさらに明らかにするために、ブタ第VIII因子との交叉反応性についても検討をおこなったが、反応性はやや低いものの共通抗原性がブタ第VIII因子C2度目にも認められた。
3.インヒビターのエピト-ブ解析:次に、イヌ第VIII因子と交叉反応性の高かったC2ドメインに着目して、インヒビターのC2の共通抗原領域をリコンビナント第VIII因子フラグメントによりアミノ酸残基2308-2312に局在化した。さらに、このアミノ酸残基を中心に15残基のアミノ酸からなるペプチドを合成して、第VIII因子 フォン・ウイルプランド因子結合、第VIII因子 リン脂質結合に対する抑制試験を行った結果、両結合を抑制する2種類の合成ペプチドを得た。現在さらに、ヒト第VIII因子インヒビターおよびイヌ第VIII因子インヒビターに対する中和実験を施行中である。
