腹膜中皮と癌細胞間での接着因子の役割と抗接着療法による腹膜転移の抑制

Research Abstract

腹膜を用いた細胞接着の新しい測定方法を確立し、これを用いて分化度の異なる2種類の胃癌細胞株の表面抗原糖鎖を検討し、ELAM-1のLigandであるSialy1 Lewis Xが腹膜接着能の低い細胞株でむしろ強く発現していることを明らかとした。この結果は、肝転移能の異なる大腸癌細胞株での検討やColon26よりクローニングして得られた低接着株での結果とも一致した。また、腹膜と癌細胞の接着をFucosy1transferaseに基質となりうるオリゴ糖や糖タンパクが特異的に阻害しうることから、腹膜上の糖転移酵素Fucosy1transferaseが、腹膜転移に関与する新たな接着因子であることを明らかにした。これは、血行性転移において既に明らかとなっているSelectinとは異なる機序により腹膜転移が形成されることを示唆している。この成果はすでに、Cancer誌に投稿し近日中に掲載される予定である。
