| Research Abstract |
アンドロゲン依存性の細胞株LNCaPおよびそのSubline(LNOおよびR)、またPC-3,TSV-PRIはいずれも高親和性のIGF-I receptorを認めたがTSV-PRIのみがIGF-Iに対して細胞増殖を認めた。conditooned medium中,IGFBP-2は、いずれの細胞株にも、その分泌を認めた。BP-3and/orBP-4は、R,PC-3ならびにTSV-PRIに分泌が認めらえた。ある種の細胞株(肺癌)では、BP-2が細胞増殖を抑制する作用があることが知られており、一方、BP-4はMCF-7細胞などで細胞増殖を促進する作用があることより、TSV-PRIにおいてはBP-4がIGF-Iの作用に促進的に働いた可能性が示唆された。いずれにせよ、BPがIGF-Iの作用を調節している可能性がある。その成果については、第5回前立腺癌ワークショップにて発表した(Effects of insulin like growth factor-I on growth of human Prostate cancer cell lines.Proceedigs of the Fifth workshop on prostate cancer)。当初IGF-Iに対してCNCaP細胞株がアンドロゲンとの併用で反応したが、そのearly orosrgeでは反応をみなかった。MTTassryの他、Thy midine incororation assayやDNA contert測定でも同様であり、その原因を検討中である。IGF-I receptorは後者も有しており、IGFBP当初の相違等を検討予定である。
