Research Abstract	前立腺癌の増殖とEpidermal growth factor(EGF)との関係については以前よりいくつかの報告がある。たとえば前立腺癌細胞株DU-145細胞で癌細胞がEGFレセプターを有し、また自らEGFを分泌することが知られている(Autocrine mechanism)。一方、最近、Insulin-like growth factor(IGF)とDU-145細胞増殖との関係が報告された。IGFによる細胞増殖機転の調節にはIGFレセプターのみならず、IGF binding protein(IGFBP)も関与しているとされる。本研究ではアンドロゲン依存性のLNCaP-FGC細胞およびそれより派生した依存性を失った細胞株(LNO,LNCaP-r)ならびにアンドロゲン非依存性細胞株TSU-PR1,PC-3を用いてIGFの細胞増殖に及ぼす影響をみた。IGF投与によりTSU-PR1では軽度ながら増殖促進がみられたが他の細胞株では増殖促進を認めなかった。LNCaP-FGCにおいてはIGFはアンドロゲン併用にても増殖効果を認めなかった。IGFレセプターはいずれの細胞株も高親和性のレセプターを有していた。IGFBPについては、BP-2をすべての細胞株に認めた。TSU-PR1,PC-3はBP-4の分泌も認めた。BP-2はヒト肺ガン由来細胞株においてIGFレセプターと競合し、IGF効果を減弱する作用があること、またBP-4にはMCF-7細胞株でIGF作用を促進させる働きが報告されている。これらの知見より、TSU-PR1はBP-4によりIGF効果が発現し、一方他の細胞株ではBP-2のレセプターとの競合阻害によりIGFが作用しなかった可能性が示唆される。いずれにせよ、これら前立腺癌細胞株の増殖の少なくとも一部はIGFにより調整されていると推定されたが、ホルモン依存性との関連は見いだせなかった。
