頭頚部癌細胞障害性T細胞クローンT細胞受容体α鎖を支配する遺伝子の塩基配列の解析

Research Abstract

下咽頭癌患者2名の腫瘍浸潤リンパ球(TIL)をInterleukin2及びirradiated autologous tumorによる定期的刺激によりlimiting dilution methodを用いて60のクローンを樹立した。このうち20は自己癌特異的で、しかもHLA-class2拘束性をしめしたので、これらについてT細胞受容体(TCR)のα鎖、β鎖の解析を進めた。TCRの解析よりVα10(11/20=55%),Vα3(6/20=30%),Vα8(5/20=25%),Vα1(4/20=20%)、Vβ8(6/20=30%),Vβ9(4/20=20%),Vβ17(4/20=20%)がdominantなusageを示した。この20のCTLのうちTCRの解析により2名の患者からそれぞれ5個、合計10個がCTLclonesと考えられた。これらのclonesのTCRのα鎖,β鎖の組み合わせが全く同一のものは存在せず、扁平上皮癌細胞上に発現させる癌抗原ペプチドは少なくとも5個以上存在すると考えられた。さらに、それらの癌抗原ペプチドのうちdominantにCTLを誘導するものは今回の実験からは否定的であった。
