Research Abstract

Hepatocyte growth factor (HGF), a hormonal mediator of liver and kidney regeneration, possesses multiple biological activities. The effect of HGF on cell growth and mitosis were investigated in the cultured rabbit corneal cells.
Primary cultured corneal epithelial, stromal and endothelial cells were prepared from Japanese white rabbits. First-passage cells were cultured in 1% Fetal BOvine Serum with 1,5 or 10 ng/ml of HGF ; 1,5 or 10 ng/ml of EGF or both factors, and observed for 96 hours. To examine DNA synthsis, three-hour bromo-deoxyuridine (BrdU) uptake was mesured in each group.
HGF stimulated growth of corneal epithelial cells and endothelial cells as a dose-dependent manner. 5ng/ml of HGF with 5ng/ml of EGF stimulated cell proliferation synergistically with greater stimulation than 5ng/ml of HGF or 5ng/ml of EGF alone. HGF also stimulated DNA synthesis significantly. But HGF had no effect on proliferation of
Marked proliferation of rabbit corneal epithelial and endothelial cells were observed with recombinant HGF.Similar growth stimulation was also found with EGF.The synergistic effect og HGF with EGF was observed on corneal proliferation and DNA synthesis in culture. Exogenous recombinant HGF may contribute to corneal epithelial resurfacing in a different manner from EGF.
