Research Abstract	ゲノムライブラリーの作成及びそのスクリーニングによる目標遺伝子の単離を目標として、研究を行なった。大腸癌細胞株WiDrからの、ゲノムライブラリーの作成に着手するのと平行して、JCRBから供与を受けたヒト末梢リンパ球ゲノムライブラリーを対象として、スクリーニングを開始した。欠失の認められる2.2kbpシグナルは、胃癌DNAをHinfI消化し、criptoのcDNAをプローブとしたサザン法にて検出されることから、ヒト末梢リンパ球ゲノムライブラリーを対象とした、スクリーニングにはcriptoのcDNAをプローブとし用いた。この結果、14の陽性クローンが得られた。これらは、制限酵素による切断パターンから6種類に分けられた。これらから、インサートを抽出し、これらをプローブとしてサザン法を施行したが、いずれのクローンからも、目的とする2.2kbpのシグナルは得られなかった。この原因としては、ライブラリーが上皮由来のものでないことや、スクリーニングの条件設定に問題があったことが考えられる。現在、WiDrからのライブラリー作成を行なっている。前年度の結果にて、2〜3kbpDNA断片からcriptoによるスクリーニングにより陽性クローンを得たが、これは部分シークエンシングにより、チトクロームp4502D6との間に相同性が認められた。同遺伝子の変化をサザン法にて検索したところ、小数例において腫瘍部での変化が認められた。しかし、その頻度は目的とする遺伝子の欠失頻度に比較し低率であった。現在、マイクロサテライト法にて、さらに、検討を行なっている。
