| Research Abstract |
種々の程度の動脈硬化組織(解剖例)および色々のタイプの乳癌組織(手術例)を採取し、Northern blotおよびin situハイブリダイゼーション法を用いて骨基質タンパクの発現を調べた。動脈硬化組織においてはオヌテオポンチンが動脈硬化の進行に伴って発現が増大し,石灰化を示す粥腫の周囲に強く発現がみられた。オステオネクチンは動脈硬化の進行に伴って発現は低下し,石灰化とオステオネクチンの発現部位には関連を認めなかった。一方、乳癌組織においては石灰化を伴う乳癌凍死部の周囲にオステオポンチンの発現がみられオステオネクチン、matrix Gla proteinは石灰化部と関係しない部に発現していた。免疫組織化学法によりオステオポンチン蛋白は石灰(リン酸カルシウム)の沈着部位と完全に一致しており、またオステオポンチン発現細胞はCD68陽性細胞すなわちマクロファージであることが示唆された。以上のことは骨の石灰化に重要な働きを持つ、骨芽細胞により産生されるオステオポンチンは、異所性の石灰沈着においてはマクロファージにより産生され異所性石灰沈着の発生に重要な役割を果たしていることを示唆している。
