| Research Abstract |
平成6年度よりSCIDマウス移植系EBV陽性リンパ腫株30例の宿主細胞へのEBV genome取り込みについての検討を行った.まず,それぞれの検体から抽出したDNAを3種類の制限酵素で切断後,3% Nusieve GM soft agar gelで電気泳導動し,1 kilo-base以下の長さのDNAを再抽出した.それをtemplateとして,6種類のAlu配列と3種類のterminal repeatのprimerを使用し,すべてのprimerの組合せでPCR増幅した.その結果,3例に400-700bpsの長さのsingle bandが検出された.直ちにsequencingを行ったところ,そのsequenceはAlu配列の部分とterminal repeatの約18塩基対までは同じ配列であったが,terminal repeat 3'側ではEBV terminal repeatにない配列であった.数回,実験を繰り返したが同様の結果であった.そのような状況で,平成6年度と7年度の半ば頃まで時間を費やしてしまった.
平成6年度はじめより,それまで研究してきたEBVの知識と技術を応用し,EBV陽性胃癌の発癌過程を解析し,Journal of Pthologyに発表した.また,EBV陽性胃癌におけるEBV genotypeの検討をし,EBVの発癌過程の解明の一助となし,現在,Journal of Pthologyに投稿中である.時を同じくして,HPVの発癌における役割についても検討を加え欧文医学誌に発表した.
