Research Abstract

平成8年度に得た成果を以下にまとめる。
1.日本側は二光束干渉法を使って卵白リゾチームの正方晶系、斜方晶系、三斜晶系結晶についての溶解度測定を行った。
2.日本側は、成長した卵白リゾチーム結晶の内部の屈折率を二光束干渉法を使って測定する手段を確立した。
3.日本側は、卵白リゾチームを液体クロマトグラフィにより精製し、高純度試料を得た。これを使って原子間力顕微鏡で結晶の成長界面の観察を行い、米国側の観察と比較し、議論を行った。
4.日本側は10Tの強磁場中で卵白リゾチーム結晶の成長実験を行い、磁場により、核形成頻度と出現する形成の配向が制御できることを明らかにした。
5.米国側はアポフェリチン、日本側はフェリチンの精製・結晶化を行い、不純物の除去がタンパク質結晶の育成に不可欠であること示した。日本側は、更にフェリチン結晶の形態相図を作製した。
6.米国側は光散乱法によりタンパク質の凝集過程について考察を行い、溶液の第二ビリアル係数を評価し、溶液内のイオン強度と分子間相互作用の関係を議論した。
7.米国側は、レーザー干渉法により卵白リゾチーム結晶の表面に形成される成長丘の形状の経時変化を精密に測定し、成長速度と微斜面の傾きの時間変化を調べた。
8.米国側は卵白リゾチーム結晶を育成する際、溶液相が二相に分離する領域があることを発見し、光散乱法によりこの相図を作製した。
9.薄い溶液層での対流の効果が無視できる厚さを評価するため、溶液の厚みと結晶のまわりの濃度分布の関係について米国側は計算機実験を、日本側は濃度分布を計測する実験を行い、対比させた。
10.日本側は高圧セル中でのタンパク質(リゾチーム)濃度のその場測定を行うとともに、結晶の成長速度の圧力依存性を調べた。
