Research Abstract	1.アフリカツメガエルならびにニワトリにおけるHGFの機能解析アフリカツメガエルならびにニワトリのHGFとc-metのcDNAクローニングを行ない、胚発生過程における遺伝子発現パターンを解析したところ、主に腹側内臓器と骨格系の形成過程において発現していることが明らかになった。さらにc-metのドミナントネガティブ体をアフリカツメガエル胚に導入し、HGFのシグナルをブロックすると、肝臓や腎臓などの内蔵臓器と骨格の形態形成に異常をきたした。これらの結果からHGfは肝臓や腎臓などの内蔵臓器の形態形成ならびに胚の骨格形成に重要な役割を担うと考えられる。 2.器官形成・形態形成におけるHGFの機能解析ラット胎児肺ではHGFとそのレセプターであるc-Metの遺伝子発現はそれぞれ間葉組織および気管支上皮組織に限局して認められ、肺原基の器官培養系においてHGFは上皮組織の分枝管腔構造形成を促進した。肺原基より間葉組織を取り除き上皮単独培養を行なったところ、HGFは肺上皮組織の形態を維持、細胞死を抑制するとともにaFGF(またはKGF)と同時に添加することにより上皮組織の細気管支様の枝分れおよびその伸長を有意に促進した。また肺原基の器管培養系にHGFの中和抗体をKGFレセプター(KGF-R)のアンチセンスDNAと組み合わせて添加することにより肺原基の分枝管腔構造形成はほぼ完全に阻害された。また、歯胚形成過程においてもHGFの発現は歯胚間葉組織にc-Met/HGFレセプターは上皮組織に局在した。歯胚の発生、形態形成におけるHGFの機能を明らかにするため、HGFのアンチセンスDNAを歯胚器官培養系に添加したところ、著しい形態形成の異常が観察された。まだ乳腺上皮組織に由来する培養細胞株に対しても、HGFはコラーゲンゲル内で枝分れした管腔構造を誘導知、HGFは様々な上皮系細胞形態形成因子であることが明らかになった。以上の結果から、HGFは上皮-間葉相互作用のメディエーターとして、肝臓、腎臓、肺、歯、乳腺など様々な器官の形成、形態形成を制御する因子であることが明らかになった。
