Research Abstract	サブテーマi)試料調整の共通化,ii)細胞内メッセンジャー,iii)CIチャネルとKチャネル,iv)Na/H,Cl重炭酸対向輸送体,v)Na/K/2Cl共輸送体,vi)Na/KATPaseとエネルギー供給)について実施。1)生理研では、新たに耳下腺の潅流実験系を開発、a)水分泌速度と酸素消費測定、Na,K輸送量測定により各輸送体のエネルギー要求を解析(村上4-9月)。Stewardと共同で容積増大時のK輸送測定(12月)b)瀬尾は京都府立医科大学に転出、NMR装置を立ち上げ、重水素NMR法・マイクロイメージングを導入、上皮膜水輸送への応用を検討(村上・瀬尾12月)。c)電解質輸送量とチャネル活性の相関を解析(石川4-6月丸中1-2月)。2)水分分泌及び開口分泌に伴う細胞間隙構造の変化をレーザー顕微鏡及びSEMにて観測、isotroterenol刺激とcholinergic刺激においてmicrofilamentによる制御様式の違いを発見(瀬川,Riva12月)。3)Purin受容体活性化による水輸送の抑制を観察(村上Soltoff.1-3月)。HPLC法にてイノシトール燐化合物の動態と輸送体燐酸化の関係を解析、purin受容体活性化によるIP3放出ブロックを発見(Dissing11月)。4)cyclicAMPによる開口分泌に関与する顆粒膜側タンパクとして新たにVAMP-2を同定、VAMP-2の標的タンパクの候補としてsyntaxin4,SNAP-23の役割を分子生物学的手法で解析(杉谷)。アミラーゼ分泌を観察、フォルボールエステルがCa依存性刺激薬による分泌増強を解析(Yoshimura4-11月)。5)シドニー大学において羊口腔腺の潅流実験を継続(村上・瀬尾10月)、導管のNaチャネル活性制御機構としてG蛋白の役割を解析(Young,Cook,Dinudom11月)。6)アンモニウムパルス法によりNa/K/2Cl共輸送体制御を解析、新しい共輸送体の働きとして陰イオンの排出機能を提案(Tumer,Evans10-11月)。7)10-11月岡崎にて研究打ち合わせ(Steward,Seo,Cook,Dinudom,Evans,Soltoff,Dissing,Tandler)、輸送体の新しい機能とプリン受容体活性化による細胞内情報伝達の制御、単一細胞と組織構築を持った細胞の容積増大の違い、開口分泌の分子機序、などについて討論、平成9年2-3月に村上・杉谷は北米にて、瀬尾はオランダにて個別に実験打ち合わせを行った。
