骨髄ミクロキメリズムによる抗原特異的免疫寛容の導入

Research Abstract	昨年度までに我々は骨髄キメラマウスにおいて、そのキメリズムが30%あればドナーに対する特異的な免疫寛容が誘導できることを示し、ドナー特異的免疫寛容誘導における部分キメリズムの有用性を実験的に証明することができた.一方でピッツバーグ大学移植外科のStarzl教授のグループは肝臓移植後の患者を詳しく解析し、拒絶反応を示さない患者では血液細胞の一部がドナー由来細胞で置き代わっていることを報告し、これがドナーに対する免疫寛容の誘導に関与している可能性を示した.我々は肝臓移植後の患者における骨髄キメラが移植肝臓中に存在する造血幹細胞によるものではないかと考え、マウスをモデルとして肝臓中での造血幹細胞の存在について検討を行った. その結果、adultマウス肝臓中にも骨髄中の造血幹細胞と同じc-Kit+Sca-1+Lin-の表現系を持つ細胞が存在することが明らかになった.この細胞は骨髄中の造血幹細胞と同様にin vitro、in vivoでコロニーを形成するだけでなく僅か500個を致死量放射線照射したマウスに移植するだけで全ての血球細胞を長期にわたり供給することが示された.これらの結果はStarzlらの報告した肝臓移植後のドナー血球による血液キメラが、移植した肝臓中に含まれていた造血幹細胞によるものである可能性を強く示唆しており、肝臓移植後のドナー特異的寛容の導入に骨髄部分キメリズムが重要な役割を果たしているという説が裏付けられた.
