| Research Abstract |
エイズ病態の免疫学的基盤に関する研究を行い、以下の研究成果を得た。1. HIV gp120のV3ループによるIL-2依存性T細胞増殖抑制(G1期停止)は、cyclin E発現抑制とcyclin E/Cdk2キナーゼ活性低下によってRbの高リン酸化が阻止されるためである。2. HIV感染者リンパ球のアポトーシスが抗Fsa抗体、抗FasL抗体で阻止され、FAS/FasL系の関与が明らかにされた。3. HIV pg160特異的T細胞レセプタートランスジェニックマウスが樹立された。これは、遺伝子治療の基礎的研究に有用である。4. HIV蛋白断片、ことにその最小認識エピトープそのものが、HIV特異的CD8^+キラーT細胞上のクラスIMHC分子に結合することによって、一時的なCTL活性の抑制が誘発される現象を発見した。この現象は、キラーT細胞が存在するにもかかわらず持続感染が成立する機構を説明しうる。5. LPS依存性無細胞NF-κB活性化系を樹立し、NF-κB活性化に関与しているIκBαリン酸化酵素の性状を明らかにした。これは、HIV感染マクロファージでみられるIL-8遺伝子転写活性低下機構解明に有用な成果である。6.エイズリンパ種ではEBVLMP1が腫瘍細胞の細胞膜に例外なく発現し、それが抗腫瘍療法の標的になることを示した。7. HIV感染症におけるヒトメモリーT細胞抗原分子の役割を明らかにする目的で、CD29由来のシグナル伝達機構を解析し、pp125FAKとパキシクリンが直接結合してT細胞の活性化に重要な役割を果たしていることを明らかにした。
