| Research Abstract |
1)アサガオからLDcDNAを単離するために、シロイヌナズナLD遺伝子のうち、上記ペプチドの繰り返し構造をコードする部分をPCRにより単離した。そのPCRに用いたプライマーでアサガオDNAを鋳型としてPCRを行ったところ、シロイヌナズナと同一のDNA断片が増幅されたので、その塩基配列を決定したところ、上記繰り返し部分のアミノ酸配列はアラビドプシスと完全に一致することが明らかになった。現在アサガオから、全長のcDNAを単離する作業を進めている。
2)アサガオのLDcDNAから明らかになったLDタンパク質のうち、抗原決定部位を推定し、そのペプチド構造を基にして抗体を作成している。
3)QPVNGに特異的な抗体を作成した。現在アサガオ・シロイヌナズナの花芽形成時期にそのペプチドが遊離しているかどうかを検索中である。
4)平成7年度の公募研究によって、LDcDNAのうち、上記ペプチドの繰り返し構造部分は保存性が高いことが明らかになったので、さらに多くの植物からLD遺伝子を単離しその構造を決定し、この遺伝子の高等植物界における分布と変異の度合いを明らかにする。
