Research Abstract

我々は精細胞の分化メカニズムを理解するため、精巣分化時期に特異的発現をする遺伝子を順次クローニングし、その機能について解析を加えることで精子形成過程を解析している。さらに、それらの遺伝子をノックアウトしたマウスを作成し、それらがコードする蛋白質の生体内機能についての解析を行っている。まず、パキテン期精母細胞から初期精子細胞までの精細胞を特異的に認識するモノクローナル抗体(TRA369)を分離した。この抗体を用いてこの分化段階にのみ発現が認められるcalmegin(精細胞分化時期特異抗原)のcDNAを分離し、その特異的発現を制御する遺伝子領域の解析をトランスジェニックマウスを作成することによって行った。さらにこのcalmegin遺伝子を欠損したマウスを作成した。この変異マウスのヘテロ同士の掛け合わせによりホモ欠損マウスを得、その表現型について解析中である。既に、このホモ欠損オスマウスは、見掛け上は正常な精子を形成するにも関わらず不妊であることを確認しており、その詳細についても検討中である。
