Research Abstract

木全は、コンドロイチン硫酸プロテオグリカンの抗一接着活性に非感受性の培養細胞2種類を見出した。昨年度、抗接着活性部位であるコンドロイチン硫酸鎖部分の細胞表面レセプター候補分子として同定したannexine VIを欠損している培養細胞を見出し、抗一接着活性におけるannexinの関連を調べる実験系を確立した。これらの細胞にannexine VI遺伝子を導入し、影響を解析している。永田は、HSP47のアンチセンスRNAの安定発現細胞株では基質であるコラーゲンの合成量の低下とともに不溶化が見られ、シャペロンとしての機能を明らかにした。北川は、抗一接着活性をもつラミニンの形成機構について、変異型ラミニン鎖遺伝子を動物培養細胞に発現させてその影響を解析した。坂倉は、テネイシン産生能を持つものと持たない乳癌細胞について細胞形態と遊走力を比較し、前者は丸く、早く遊走し、癌浸潤能が高いことを見出し、その抗一接着活性と癌転移との関係を明らかにした。小原は、テネイシンの示すcrypticな細胞接着活性の発現にはRGD非依存性インテグリンの関与があることを見出して、抗一接着活性との相違を明らかにした。大平は、神経回路網形成期のラット脳に新規の膜結合型プロテオグリカンを見つけニューログリカンCと名付け、細胞接着における影響から神経突起形成における役割を示唆した。岡山は、低転移肺癌細胞と高転移株について、前者は高いシンデカン-2発現によりストレスファイバーの形成が、後者ではインテグリンのみによりラフリング膜形成が誘導される。この細胞接着の差が細胞増殖、運動能の相違をもたらす可能性を示した。関口は、フィブロネクチンのインテグリン結合活性が、特定領域の選択的スプライシングにより制御されていることを発見し、スプライシングの意義を明らかにした。野田は、チロシンフォスファターゼ活性をもつ脳プロテオグリカンを同定し、中枢神経細胞には抗一接着活性を示すことを見いだした。そのリガンドの解析により、プレイオロフィン受容体として機能していることが示された。祖父江は、ラミニンの平滑筋細胞分化因子としての機能を示すとともに、IGF-Iが必須であることが明らかにした。ラミニン接着とIGF-Iを介する分化維持シグナル系としてチロシンリン酸化の関与を見い出した。稲垣は、リン酸化細胞骨格蛋白質特異抗体群の作成に成功して分裂細胞におけるこれらのリン酸化を解析した。Rho制御性キナーゼに類似した未知のキナーゼ(CFキナーゼ)により分裂後期に分裂溝の中間径フィラメント構成蛋白質がリン酸化されることを示し、細胞接着との関連を示唆した。月田は、カドヘリン結合蛋白質でもあり、シグナル伝達因子でもあるβカテニンを安定化させる方法を見出し、さらにそれを核内に発現させると、インテグリン系の活性化が引き起こされることを見出した。第2回目の班会議を名古屋で開催し、班員相互の交流と情報交換を行った。
