ムチンによる細胞交通と病態の制御

Research Abstract

ムチンは、細胞がその表面に特定の構造の糖鎖を高密度に多数発現する必要があるときに用いられる分子で、糖含量が高く非常に分子量の大きい糖蛋白質である。ほとんどの糖鎖はいわゆるムチン型のO-linkedのものであり、これらの糖鎖の結合するペプチド部分は細胞外に存在ししばしば繰り返し構造を持つ。最近の研究結果から、細胞どうしの認識とそれによる活性化、さらには体内分布を決める細胞交通を制御する分子がムチンであることが明らかになった。特に、免疫細胞と上皮細胞(特に癌化したもの)などが関与する病態形成において重要性が強く示唆された。本年度は、ムチンのグリコシレーションとこれを認識する分子であるレクチンに関する次のような成果を得た。(1)硫酸化ルイスa糖鎖を含むムチンがマウスの消化管(大腸の遠位)に発現する事を明らかにした。(2)ヒト大腸癌細胞表面の肝臓に発現する接着分子に結合すると考えられるムチン糖鎖の構造を明らかにした。糖鎖の還元末端附近にはコア2構造を持つポリラクトサミンで、フコースは分岐近くのN-アセチルグルコサミンにひとつ付着していた。(3)MUC2ムチンのタンデムリピートの一部をなすスレオニンが三回繰り返す構造が、異なるペプチドN-アセチルガラクトサミニルトランスフェラーゼ(GalNAc-T)によってどのようにN-アセチルガラクトサミンの転移を受けるかを査定した。GalNAc-TlはまずN-末端方向から1番目次いで3番目のスレオニンに、GalNAc-T2は1番目のスレオニンのみに、GalNAc-T3はまず3番目、次いで2番目、次いで1番目の順で全てのスレオニンにGalNAcを転移することが明かとなった。すなわちムチンの糖鎖のアレンジが異なるGalNAc-Tによって実現することが明かとなった。(4)ヒトMUC1ムチンのcDNAベクターでマウスを免疫することによって、ヒトMUC1ムチンに対する免疫応答が誘導され、MUC1ムチンを発現したメラノーマの実験的な肺転移が抑制されることが明かとなった。
