| Research Abstract |
1.胃癌キラーT細胞(CTL)のT細胞レセプター(TCR)遺伝子解析
ヒト胃癌HST-2に特異的に反応するCTLはTCRVα7、Vβ20ファミリーを使用していることがわかった。一方、胃癌SSTWのCTLTCRも同じファミリーを使用していた。しかし、各々のCDR3の構造を調べるとN-diversityは明らかに異なり、この部分で特異的認識を行っていることが推測された。また、HST-2の系で、このCDR3をもつT細胞のclonal dominanceをみたところ、いかなる病期から得たT細胞CDR3は同じ構造を示していた。このことは、このCDR3をもつT細胞がclonal dominanceをもつことを意味し、このことは対応するHST-2の抗原もmajorな単一のものが存在することを示唆した。
2.HST-2の拒絶抗原解析
酸油出法により、HLA-A31に提示される10ヶのアミノ酸よりなる抗原ペプチドを同定した。この抗原は他の胃癌にも発現が示唆され、これらをCTL TCRが認識していると考えられた。
