生殖腺附属器官における細胞増殖および細胞死機構の解析

Research Abstract

雄マウスの精巣を外科的に腹腔内につり上げ,精嚢の重量,DNA断片化,3'-エンドラベリング法によるアポトーシス細胞の定量化を行うとともに,mRNAを抽出し,RT-PCR法によりTGF-β1.2.3,TNF-α,TNF receptor,Fas,Fas-ligand,KGF,androgen receptor,cathepsion D,SGP-2等の発現を調べた。この結果,潜伏精巣により1日後に精巣重量の減少,DNA断片化が認められ,アポトーシス細胞の率が7倍に増加した。これに伴い,TNF-α receptor,TGF-β2,3,FasのmRNA発現が増加した。しかし,機能的なFas(lpr,lprcg)あるいはFas-ligand(gld)をもたない、あるいはその両方を持たないマウス(gld/lpr)でも潜伏精巣により精子細胞の細胞死が誘導された。また,離乳後の乳腺の退縮機構を同様に解析した結果,離乳1日後にはTGF-βs,TNF-αのmRNA発現が増加した。P53ノックアウトマウスから乳腺を摘出し,10%血清を含む培養液で細胞培養し,抗Fas抗体およびTNF-αを添加したところ,抗Fas抗体では24時間以内に60%以上の細胞が死に,TNF-α単独では細胞死は誘導しなかった。しかし,アクチノマイシンDの共存下では,抗Fas抗体および,TNF-αは約80%以上の細胞を殺した。また,機能的なFas(lpr,lprcg)あるいはFas-ligand(gld)をもたない、あるいはその両方を持たないマウス(gld/lpr)でも離乳により、乳腺の細胞死が誘導された。以上の結果から,Fas系およびTNF-α系が乳腺の細胞死に独立に関与していることが明らかとなった。さらに,雌マウス生殖腺附属器官,妊娠依存性乳腺腫瘍、脱落膜のアポトーシス機構も調べている。
