Research Abstract

The carp hematopoietic cells of pronephros can proliferate and form colonies in the presence of homologous carp serum. After 3-5 days of culture, colonies consisting of small round cells were observed. The number of cells in the colonies increased steadily. Colonies were formed on an adherent cell layr. The proliferated cells in the colonies were separated from the colony into the culture medium. These floating cells were constantly produced for more than one month. Morphologically, the majority of the floating cells had features of immature type of cells which were mainly round cells with basophilic cytoplasm and a round nucleus. Hidrocortisone enhanced the growth of granulocytes, whereas B lymphocytes proliferated predominantly in the absence of this hormone. Such a method contributes to the investigation of the roles of fish polymorpho-nuclear phagocytes and lymphocytes in host defense. The cells had phagocytotic activity against Latex beads and had acid phosphatase and nonspecific esterase activities. Electron microscopically, the cells had slender microvilli, micropinocytotic vesicle-like vacuoles, and microfibrils, similar to red sea bream resident peritoncal macrophages. The cells had receptors for red sea bream IgM on cell surface. The cultured cells migrated to FMLP and the supernatants of the cultured cells. The results suggest that resident peritoneal macrophages produce and release chemotactic factor-line substances in abdominal cavity. In a colonial ascidian, physical stimulations induce strong luminescence in the tunic. Three types of tunice cells are morphologically discriminated as morula-like tunic cells, tunic phagocytes, and tunic granulocytes. Tunic phagocytes contain round granules, clear vacuoles, and occasionally phagosomes. Tunic granulocytes characterized by a number of elliptical granules and they occasionally contain phagosomes and round granules that are similar in structure to tunic phagocytes. Less
