| Research Abstract |
未分化大細胞型リンパ腫は、ヒト悪性リンパ腫の2-7%を占める悪性リンパ腫-亜型である。その発生機序は不明であったが、我々は最近、この型のリンパ腫の40%において、染色体2番と5番の転座によって2つの遺伝子ALKとNucleophosminが融合して新種のキメラ遺伝子が生じており、その遺伝子の産物であるp80が高発現していることを見いだした。本年度われわれはこのキメラ蛋白質p80について、その腫瘍細胞内でのシグナル伝達機序を検索し、それが正常細胞の増殖シグナルの伝達経路の特定部位に直接に働いてこれを刺激していることを明らかにし、またこの発現それ自体で細胞の増殖促進が著しく高まることを明らかにした。またそのキメラ遺伝子の片側の構成成分である機能不明の新種遺伝子であるALK遺伝子について、これが正常脳に多量に発現しており、神経の機能と関連していることが推定されることを示した。さらにp80の発現の有無によって、従来鑑別診断の困難であった未分化大細胞型リンパ腫とホジキン病が区別できること、すなわち前者の多くにはこの蛋白質が高発現しているが、後者にはこれを発現しているものがほとんどないことを、欧米の諸グループとの共同研究で明らかにした。これらの知見は、未分化大細胞型リンパ腫は発生機序の単一な悪性リンパ腫亜型であることを示していると判断される。
