心肥大に関与する遺伝子群の同定

Research Abstract

心臓レニン・アンジオテンシン系に関する検討。
心室組織内で発現しているACEが、組織内のアンジオテンシンIIの生成量を規定している事を、aorto-venous fistulaによる心肥大モデルを用いて明らかにする事ができた。また、心筋梗塞といった、特殊な病態下では、組織に浸潤してくるマクロファージが、レニンを発現する事を見出した。
Differntial screeningによる肥大に関係する遺伝子群の同定。
全部で、8種の遺伝子を同定したが、その多くは、extracellular matrixに対応する遺伝子であり、心肥大、心不全に於ける問質の線維化の重要さが浮き彫りになった。
F2ラットを用いた遺伝学的解析。
ラット染色体1番及び13番に心肥大と相関する遺伝座位を見出した。一つは、血圧にも影響を与えるが、他方は、血圧には無関係である。現在座位をもう少し絞り込んでおり、また、座位とbiochemicalなマーカーの相関関係を見出す努力をしている。
