Research Abstract	粥状動脈硬化症の形成には大きく二つの現象が関与する。血管壁におけるコレステロールエステルの蓄積と、血管壁における細胞成分の病的反応である。私共は細胞内コレステロール代謝調節という観点より、新しい粥状動脈硬化症の内科的治療法の開発に発展したい。そこで、細胞内コレステロール代謝調節に関与するACAT及びNCEHのgene targettingを行うことにより、血管壁コレステロール蓄積機構を明らかにしようとしている。またホルモン感受性リパーゼのgene targettingを行うことにより、本酵素とNCEHの同一性、即ち脂肪細胞での脂肪分解と血管壁でのコレステロールエステル分解反応のクロスオーバーについて、遺伝子レベルで明確にしようとしている。既にACAT及びホルモン感受性リパーゼのcDNAクローニングを終了し、gene targettingを開始している。gene constractを作製して、ES cellに導入して、マウスへの遺伝子導入を試みている段階である。本研究を通じて、細胞内コレステロール代謝調節の異常や、随伴する血管壁における細胞成分の病的反応を明らかにして、粥状動脈硬化症の成因を解明していきたい。
