Research Abstract	1)Asp1179変異,およびLeu1193変異インスリン受容体はパルスラベル法により,変異受容体のプロレセプターに明らかな消失速度の亢進がみとめられた。小胞体からゴルジ装置に達するまでの段階でプロセッシング障害を生じてプロレセプターが蓄積するΔexon13変異受容体に,Asp1179変異,あるいはLeu1193変異を合わせ持った重複変異受容体では分解が早く正常2.5倍の分解亢進をみとめた。 2)これらの変異変容体の分解亢進を生じる機序として,細胞質に存在する分子シャペロンとの関連を検討した。両変異受容体はHsp90と強く結合し、Hsc70,Hsp90両方の存在下ではHsp90とさらに強い結合を示した。 3)分子シャペロンHsp90が変異受容体の分解亢進に実際に関与しているかを検討するため,Hsp90抗体を正常、あるいは変異受容体を発現させたCOS7細胞に微量注入した。抗体の注入により、変異受容体の発現が強くなることが明らかとなり、Hsp90の機能の抑制により、変異受容体の分解が抑えられたことが示唆された。
