| Research Abstract |
Free radical familyはいろいろな病態に関与していることが知られているが,関節症における報告はない.我々は間接破壊にfree radical(superoxideおよびNO)の関与を示唆する結果を得た(Agents Actions,1994).本研究の目的は,1)軟骨細胞におけるfree radicalの産生機序,2)軟骨代謝に対する影響,3)臨床上の意義を検討することにある.平成7年度の目標として,1)関節軟骨よりのsuperoxide産生の検討を挙げたが,科学研究費補助金により購入したArgus50を用いることによりこれに成功した.また2)薬剤添加時のfree radicalの誘導も目的の一つであったが,これについても軟骨の層別解析を行い,IL-1添加時のNO産生に差のあることを見い出した(Inflammation Res,1995).さらに3)その条件における軟骨代謝の変化も目標の一つとしたが,これについてもウシ軟骨細胞にIL-1を添加することで関節破壊を再現することに成功した(J Pharmacol Exp Therap,in press).
以上の結果より,当初平成7年度の目標として設定した項目については全て達成できており,研究は順調に進捗していると言える.平成8年度はこれらの見知の上にたって,関節症への関与を検討する予定である。
